It’s generally recommended to add both grandparents and the mother to a grandparent DNA test if possible.  Every person inherits half of their DNA from their mother and half from their father. When the mother is tested, her DNA is eliminated from the child's genetic profile. The remaining unmatched DNA is then known to be that of the biological father.

Furthermore, if both grandparents participate then the entire possible genetic makeup of the alleged father is represented in the test.  This greatly increases the conclusiveness.  A grandparent DNA test however, can still be performed with only one alleged grandparent and the child.
