A Sibship Analysis Can Help Answer 3 Questions:

  • Do the Siblings share any common parent? (Are they brother or sister at all?)
  • Do the Siblings share one parent? (Are they half siblings? half brother - half sister)
  • Do the Siblings share both parents? (Are they full siblings? full brother - full sister)

Basically, every individual is has 23 pairs of homologous chromosomes. Seperately, there are a total of 46 chromosomes. Of those chromosomes 22 pairs are autosomes, (which are provided half by the mother and half by the father, except the sex chromosomes in which the mother always provides the X while the father may provide and X or Y chromosome. This last paired chromosome is what determines the sex of the person ( XX if a female or XY if a male).  In a scenario where two persons are trying to determine if they share a common father, if the mother is analyzed along with the alleged brother/sister, the known parent (mother in this case) would be compared to her child(ren) and thus the genes contributed by the mother would be identified and separated out of the sibling's DNA profile.  The remaining genes are from the biological father, and can be more easily compared to one another determining if the siblings are half siblings, full siblings, and/or not even related at all. A sibling DNA test will help answer questions regarding your paternity. By testing against a sibling paternity testing does not need to be performed.

Understanding The Test Process:

First, home DNA testing can provide conclusive answers & the DNA collection process is quick, easy, and painless. As part of the half sibling dna testing process, our laboratory will analyze the chromosomes for each person.  Based on the genetics, a siblingship index is determined. If the index is less than 1.00, this indicates that a biological relationship is less likely.  If the index is greater than 1.00, this indicates that the two tested individuals are more likely to be biological brother/sister. To analyze the possibility that the brothers/sisters share only one common parent versus two common parents (mother or father), a half sibling index is obtained. The ratio of the half sibling index versus the full sibling index would provide you with an idea of whether the two individuals are more likely be half siblings sharing only one common parent (e.g. mother only) or full siblings and sharing two common parents (mother and father).

Each person’s DNA is as unique as their fingerprint.  When the issue of siblingship is in question, the recommended method is to analyze the parents (mom) along with the alleged brother/sister. By eliminating mom's DNA from the sibling's profile, you can positively identify the remaining genes as that of the biological father. In situations where the alleged parents (mom) are unavailable, a high sibling DNA testing accuracy may still be achieved.
